چکیده

Estrogenic regulation of gene expression involves interaction of the hormonewith its receptors, which undergostructuraland conformational changes to interact with specific DNA sequences. Putrescine, spermidine, and spermine, are ubiquitous cellular components. We studied the effects of these polyamines on rabbit uterine estrogen receptors by sucrose gradient centrifugation and ligand dissociation kinetics. The native 7S receptor converted to a 9S-10S form in the presence of 100 ¿IMspermi dine or spermine. Higher concentrations caused precipitation of the receptor. This precipitation was reduced by RNase treatment of the receptor. RNase-treated receptors sedimented at 4S and 7S regions of sucrose gradient. The dissociation rate constant (k) of the 4S receptor is 2.8 x 10 ' min ' in the presence of 1 mM spermidine, compared to a control value of 7.7 x IO"3min"'. Similar effects were observed with putrescine and spermine. The dissociation of the RNase-treated 7S receptor was biphasic, with about 50% of the receptors dissociating at a faster rate (A, = 40 x 10~3 min'1) than the other half (k2 = 7.4 x IO'3 min"'). Spermidine (1 HIM)caused a 2-fold reduction in ¿2, whereas k, was not affected. This study shows that polyamines affect the structural organization and ligand dissociation kinetics of estrogen-receptor com plexes.

Effect of Estrogen Progesterone LH and FSH on Oxidant and Antioxidant Parameters in Rat Uterine Tissue

Background To attain whether the effects of low and high doses of estrogen progesterone luteinizing hormone (LH) and follicle stimulating hormone (FSH) in acute and chronic administration were related to oxidant and antioxidant parameters in rat uterine tissue as well as cyclooxygenase-1 (COX-1) activity. MaterialsAndMethods Acute and chronic administration of estrogen (1 and 5 mg/ kg) progeste...

Self-assembly of an oligodeoxyribonucleotide harboring the estrogen response element in the presence of polyamines: ionic, structural, and DNA sequence specificity effects.

Estrogenic regulation of gene expression is mediated by the binding of the hormone to its specific receptor, estrogen receptor (ER), which undergoes structural and conformational alterations to recognize specific DNA sequences, estrogen response elements (ERE), in responsive genes to trigger a series of events culminating in the transcription of these genes. Polyamines are ubiquitous cellular c...

Rabbit myometrial adrenergic sensitivity is increased by estrogen but is independent of changes in alpha adrenoceptor concentration.

The ability of estrogen to increase the alpha-1 adrenergic sensitivity of rabbit uterine smooth muscle was compared with its effects on the concentration of alpha adrenoceptor subtypes in myometrium. Compared to ovariectomized controls, estrogen treatment was found to increase the adrenergic contractile sensitivity of rabbit uterus determined in vitro. Estrogen treatment increased uterine alpha...

Effects of natural and synthetic polyamines on the conformation of an oligodeoxyribonucleotide with the estrogen response element.

We studied the effects of natural and synthetic polyamines on the conformation of an oligodeoxyribonucleotide (ODN1) harboring the estrogen response element (ERE) by circular dichroism (CD) spectroscopy and polyacrylamide gel electrophoresis. Putrescine and spermidine had no marked effect on the CD spectrum of ODN1. In contrast, spermine provoked and stabilized two characteristic changes in the...

Regulation of rabbit myometrial alpha adrenergic receptors by estrogen and progesterone.

The effects of estrogen and progesterone on uterine alpha-adrenergic receptors were investigated by direct receptor-binding studies. Immature female rabbits were primed with estrogen by intramuscular injections for 4 days. Other rabbits were primed with progesterone by injections of estrogen for 4 days followed by injections of progesterone for 4 days. The alpha adrenergic antagonist, [3H]dihyd...
